Investing.com -- Shares of Arrowhead Pharmaceuticals (NASDAQ: ARWR) leaped 6.7% on Wednesday after the biotechnology firm unveiled highly anticipated clinical data that could reshape the landscape of Alzheimer's treatment. In a landmark achievement for RNA therapies, the company's experimental drug ARO-MAPT demonstrated an unprecedented ability to reach the brain and drastically reduce the proteins associated with neurodegeneration.
The interim Phase 1/2a data revealed that ARO-MAPT achieved a staggering 73% average reduction in total tau proteins in the cerebrospinal fluid of healthy volunteers by Day 105, with some individuals seeing drops as high as 85%. This milestone represents the medical community's first successful clinical validation of a platform capable of delivering short interfering RNA (siRNA) across the notoriously impenetrable blood-brain barrier to achieve significant gene silencing directly within the central nervous system.
Designed to combat Alzheimer's disease and other debilitating tauopathies, ARO-MAPT works by shutting down the expression of the tau gene before the resulting proteins can accumulate and damage brain cells. The trial tested multiple regimens, with the highest dose of 12.8 mg/kg administered just twice—on Days 1 and 15—yielding the most dramatic 73% reduction. Even at lower doses, the therapy proved highly effective, producing mean reductions ranging from 40% to 69%.
Beyond the sheer efficacy of the drug, the longevity of its effects offers a massive quality-of-life advantage for future patients. Because the profound tau reduction was sustained a full 90 days after the final dose, Arrowhead anticipates the therapy could eventually be administered just four times a year, or potentially even less frequently.
Crucially, the groundbreaking treatment has proven highly tolerable so far. Investigators reported no serious adverse events, deaths, or trial discontinuations. The side effects that did occur were overwhelmingly mild, limited primarily to injection-site reactions, minor bruising, and routine headaches.
Armed with this robust safety and efficacy profile, Arrowhead is now pushing forward to complete its multiple-ascending-dose trials in healthy volunteers while aggressively enrolling patients suffering from early-stage Alzheimer's disease. The broader medical and investment communities will be watching closely as the next major clinical updates are slated for 2027.
