Propanc releases preclinical PDAC data for PRP, eyes 2027 trial

September 22, 2026 8:31 AM EDT

Propanc Biopharma (Nasdaq: PPCB) released a comparative analysis of its lead drug candidate PRP against clinical data from Erasca's pan-RAS molecular glue ERAS-0015 in pancreatic ductal adenocarcinoma (PDAC), according to a company statement.

In orthotopic and patient-derived xenograft preclinical models of advanced PDAC, PRP — a fixed-ratio combination of the pancreatic proenzymes trypsinogen and chymotrypsinogen — achieved greater than 90% mean tumor-growth inhibition versus vehicle controls and a median overall survival extension of more than 2.5-fold compared with controls. The preclinical data also showed reduced metastatic burden in the liver and peritoneum, tumor microenvironment remodeling, and enhanced sensitivity of chemo-resistant PDAC cells to gemcitabine and nab-paclitaxel.

PRP holds FDA Orphan Drug Designation for pancreatic cancer and is not restricted to a specific RAS genotype. The company said its mechanism — promoting differentiation of malignant cells, reversing epithelial-mesenchymal transition, and depleting cancer stem cells — differs from RAS inhibition and is intended to complement rather than compete with RAS-targeted therapies.

Erasca's ERAS-0015, which received FDA Fast Track designation for metastatic pancreatic adenocarcinoma on August 24, 2026, reported unconfirmed overall response rates of 40% to 42% at pharmacologically active doses and a 57% unconfirmed 8-week overall response rate at its 32 mg recommended expansion dose in second-line or later KRAS G12X PDAC patients in Phase 1 trials.

The Propanc analysis also references Revolution Medicines' daraxonrasib, which received FDA approval in August 2026 for pretreated metastatic PDAC.

Propanc said it is progressing GMP manufacturing and clinical partnerships in support of a planned Phase 1b first-in-human multicenter, open-label study targeting approximately 40 to 50 patients with advanced solid tumors, including pancreatic, ovarian, and other refractory cancers. First patient dosing is targeted for February 2027, with a clinical trial application expected in the coming months.
