C4 Therapeutics sets cemsidomide trial timelines through 2028

C4 Therapeutics Inc. (NASDAQ: CCCC) outlined development milestones for its cemsidomide drug candidate, including plans to initiate a Phase 2 trial in the first quarter of 2026 and a combination study in the second quarter.

The company will begin the Phase 2 MOMENTUM trial of cemsidomide and dexamethasone in Q1 2026, with enrollment expected to complete within 12 months. The recommended Phase 2 dose is 100 micrograms, selected after discussions with the FDA.

C4 Therapeutics plans to start a Phase 1b trial combining cemsidomide with elranatamab in Q2 2026. The company expects to present initial overall response rate data for the MOMENTUM trial in the second half of 2027 and submit a new drug application by year-end 2028.

Cemsidomide is an investigational oral molecular glue degrader targeting IKZF1/3 transcription factors in multiple myeloma treatment. Phase 1 trial data showed the two highest dose levels achieved 40% and 53% overall response rates respectively in heavily pretreated patients.

The company reported completing enrollment in its Phase 1 cemsidomide trial during 2025 and developing a regulatory pathway that could lead to two potential accelerated approvals - one for fourth-line treatment and another for second-line combination therapy.

C4 Therapeutics stated it has cash runway through the end of 2028. The company is also advancing internal discovery projects focused on inflammation and neurodegenerative diseases, targeting up to three investigational new drug applications by year-end 2028.

The MOMENTUM trial will enroll approximately 100 patients with relapsed/refractory multiple myeloma who have received at least three prior treatment regimens. Multiple myeloma affects about 36,000 people annually in the United States.
