study links NR supplement to reduced muscle epigenetic aging marker

Niagen Bioscience, Inc. (NASDAQ: NAGE) announced the publication of a peer-reviewed analysis in the journal Aging Cell examining the effects of its Niagen nicotinamide riboside (NR) supplement on epigenetic aging markers in skeletal muscle.

The analysis, led by researchers at the Minerva Foundation Institute for Medical Research in Helsinki, Finland, examined skeletal muscle and blood samples from three previously published clinical studies. Researchers applied seven epigenetic clocks to assess epigenetic age acceleration, a molecular measure of biological aging.

According to the press release, five months of Niagen NR supplementation at 1,000 mg per day was associated with a reduction in muscle epigenetic age acceleration of approximately 2.5 years, as measured by the muscle-specific MEAT epigenetic clock. Significant reductions were also observed using the DunedinPACE and PCHannum clocks. One clock, PCGrimAge, showed an increase in epigenetic age acceleration following supplementation.

The study also found a correlation between changes in mitochondrial DNA content and changes in MEAT-derived epigenetic age acceleration, suggesting a potential relationship between mitochondrial adaptations and epigenetic clock measures.

In laboratory experiments using human muscle cells, Niagen NR increased NAD+ levels by 1.52-fold compared to control cells, though no significant changes in epigenetic aging markers were observed in that cell model.

"This is among the first human studies suggesting that Niagen NR supplementation may alter epigenetic aging in skeletal muscle, rather than simply increasing NAD+ or improving mitochondrial function," said Rob Fried, CEO of Niagen Bioscience.

Senior study author Dr. Eija Pirinen of the University of Helsinki and University of Oulu noted that "additional research must be conducted."

The company acknowledged in its forward-looking statements that the findings are derived from a secondary and largely correlational analysis of previously conducted studies and may not be confirmed by future research.
