XtalPi enters drug development partnership worth over $400 million
XtalPi (2228.HK) announced a strategic partnership with an international biopharmaceutical company to develop small molecule therapeutics targeting a G protein-coupled receptor (GPCR). The collaboration builds on a pilot phase where XtalPi's quantum physics and AI algorithms achieved breakthrough hit rates against the metabolic target.
Under the agreement, XtalPi will receive an upfront payment and full funding for early research and development efforts. The company is eligible for preclinical, clinical, and commercial milestones, plus future royalties, bringing total potential deal value to over $400 million.
The partnership focuses on a GPCR target that exhibits conformational plasticity, making it difficult for small molecules to engage selectively. The absence of publicly available co-crystal structures of the receptor bound to small-molecule ligands creates additional challenges for traditional high-throughput screening methods.
XtalPi deployed multiscale enhanced sampling simulations to map the receptor's conformational landscape and implemented a multi-conformational screening strategy. The company used quantum physics models and AI algorithms for virtual screening across hundreds of millions of commercial compounds, applying its XFEP platform for binding affinity prediction.
For the comprehensive collaboration phase, XtalPi will deploy its structure-based drug design platform, integrating quantum physics, generative AI, and automated chemical synthesis through a Multi-Agent system to drive Design-Make-Test-Analyze cycles.

XtalPi, founded in 2015 by three MIT physicists, operates an R&D platform powered by quantum physics, artificial intelligence, and robotics for pharmaceutical and other industries.
