H.C. Wainwright analyst Andrew S. Fein reiterated a Buy rating and $22.00 price target on WAVE Life Sciences (NASDAQ: WVE).

The analyst commented: "In a catalyst-rich year, the WVE-007 data readout remains the most important event, in our opinion. With the dosing completed in the first two cohorts, INLIGHT trial will follow predefined time points of one, three, and six months, which will assess safety, tolerability, target engagement, weight loss, and biomarkers reflective of healthy weight reduction. The best-case scenario for ‘007 based on the discussion yesterday, seems like a weight loss kinetics similar to semaglutide, however, exclusive to fat rather than muscle. We think the expectation of weight loss coming from solely fat lies in the fundamental biology inherent to the inhibin Activin E signaling pathway. INHBE was identified through an unbiased human genome screen, revealing that the loss of its functional allele is linked to weight reduction, redistribution of abdominal fat, and a decreased risk of type 2 diabetes. Preclinical studies by other groups have demonstrated that inhibin knockout mice exhibit enhanced lipolysis in both epididymal and subcutaneous white adipose tissue (WAT). Additionally, Activin E has been found to be overexpressed in steatotic liver, further highlighting its role in metabolic regulation. Support for this TGF-beta pathway comes from Phase 2 clinical data on bimagrumab, as well as Pfizer’s internal research on an Activin Type II receptor antibody, which suggests that reducing ALK7 signaling may promote lipolysis by activating key lipases in adipose tissue while preserving muscle mass. The fact that the Activin E has been overexpressed in the steatotic liver also suggests the potential of pleiotropic effect of the inhibin knockdown approach. The biomarker data apart from Activin E levels might help understand the pleiotropic effect. As for safety, given the approach targets the ligand, we expect no off-target effect. Although given the pathway is novel, and preclinical data comes from the mouse model, we remain cautious till the data reads out. Overall, we think if safety is minimal, and the weight loss kinetics is similar or comparable to GLP-1R agonists, it can be encouraging. If the data also reveals signs of sustainability of weight loss we think it might be a home run for Wave."
