H.C. Wainwright Reiterates Buy Rating on Zentalis Pharmaceuticals (ZNTL)

H.C. Wainwright analyst Andrew Fein reiterated a Buy rating and $10.00 price target on Zentalis Pharmaceuticals (NASDAQ: ZNTL).

The analyst commented: "Zentalis has presented updated results from Part 1b of the ongoing DENALI Phase 2 trial, which evaluates azenosertib (400 mg QD, 5:2 schedule) as monotherapy in patients with platinum-resistant ovarian cancer (PROC). The latest data, disclosed at SGO 2025, highlights a higher objective response rate (ORR) in patients with Cyclin E1+ tumors, reinforcing the predictive value of Cyclin E1 overexpression for therapeutic benefit. Among response-evaluable Cyclin E1+ PROC patients, the ORR reached 34.9% (15/43; 95% CI: 21.0 - 50.9), while the intent-to-treat (ITT) Cyclin E1+ population had an ORR of 31.3% (15/48; 95% CI: 18.7 - 46.3) and a median duration of response (mDOR) of 6.3 months. These outcomes build upon previously reported data, where an ORR of 31.3% was noted in the CCNE1+ population, with a mDOR of approximately 5.5 months. Notably, these efficacy results compare favorably with historical chemotherapy benchmarks (4-13% ORR, 3.2-3.8 months median progression-free survival [mPFS]). A key takeaway from DENALI Part 1b is the validation of Cyclin E1 overexpression as a biomarker for azenosertib responsiveness. Zentalis estimates that approximately 50% of PROC patients exhibit Cyclin E1 overexpression, irrespective of CCNE1 gene amplification. This insight could aid in optimizing patient selection for future trials and potentially enhance treatment outcomes by targeting a molecularly defined subset of PROC patients. Reiterate Buy and our price target of $10."
