Oncolytics reports positive preclinical data for pelareorep-RAS combo
Oncolytics Biotech Inc. (NASDAQ: ONCY) announced initial preclinical study results showing that pelareorep combined with RAS inhibitor modalities demonstrated greater anti-tumor activity than individual treatments alone in a solid tumor model.
The clinical-stage company is developing pelareorep, an investigational immunotherapy delivered intravenously. The preclinical findings support further evaluation of the combination approach in cancer treatment.
Based on these results, Oncolytics plans additional studies in pancreatic ductal adenocarcinoma and colorectal cancer models. The planned research will evaluate the combinations' effects on immune activation, tumor response durability, and time-to-resistance.
The ongoing work includes evaluations of pelareorep combined with KRAS G12C inhibitors, pan-RAS inhibitors, and additional next-generation RAS pathway-targeting agents in RAS-mutated tumor models.
"We believe these initial findings further support pelareorep's potential to serve as an immune-priming backbone for next-generation targeted therapies," said Jared Kelly, Chief Executive Officer of Oncolytics.
Kelly noted that RAS-mutated tumors, particularly pancreatic and colorectal cancers, remain difficult to treat due to intrinsic immune resistance and the emergence of therapeutic resistance over time.
The company expects to present full results from the initial preclinical studies in fall or winter 2026.
Pelareorep has received Fast Track designation from the FDA for colorectal and pancreatic cancer. The company is advancing pelareorep in combination with chemotherapy and checkpoint inhibitors in metastatic gastrointestinal cancers.
